Bilirubin interference in kinetic creatinine determination.

194 CLINICAL CHEMISTRY, Vol. 25, No. 1, 1979 gas chromatography before and after acid (6 mol/L HC1) and alkaline (NaOH) hydrolysis. We frequently observe a peak in the lactic acid position after alkaline hydrolysis. After acid hydrolysis of the same urine no lactate isobserved, but a peak appears in the position of levulinic acid. The mass spectra of these two peaks were identical with those of authentic lactic and levulinic acids, respectively. On hydrolysis of urine from diabetic patients, the lactate and levulinate increased by 30 to 50 times normal. We thought that these acids were formed from glucose by degradative reactions, and confirmed this by hydrolysis of a 10 g/L solution of glucose under acid and alkaline conditions. Again, levulinic and lactic acids respectively, were found by gas chromatographymass spectrometry. Levulinic acid is not a normal constituent of human urine (6) but has been reported to be present after medication

Kinetic serum creatinine assays. II. A critical evaluation and review.
We have evaluated the Technicon AutoAnalyzer method, the Beckman Creatinine Analyzer II, DuPont aca, Electro-Nucleonics GEMSAEC, and Instrumentation Laboratory Model 916 kinetic methods for determination of serum creatinine. These methods, which involve the Jaffé reaction, all showed a statistically significant proportional error relative to an ion-exchange pretreatment method for creatinine in...
متن کاملBilirubin interference in a peroxidase-coupled procedure for creatinine eliminated by bilirubin oxidase.
A colorimetric peroxidase-coupled procedure for determination of creatinine in human serum and urine is described. A 30-s sample pre-treatment with bilirubin oxidase eliminates interference from endogenous bilirubin. The 4-aminoantipyrine-2-hydroxy-3,5-dichlorobenzenesulfonate chromogen system of this method is about fourfold more sensitive than current procedures that involve monitoring NAD(P)...
متن کاملInterferences in current methods for measurements of creatinine.
We measured the interference of carbonyl compounds, drugs, and other substances in human serum on the determination of creatinine by the two-point, fixed-time kinetic modification of the Jaffé reaction as well as by four enzymatic methods. We added known concentrations of the interfering substances to a solution of creatinine in water. For bilirubin, we used both pooled normal sera with added b...
متن کاملA solution to the problem of bilirubin interference with the kinetic Jaffé method for serum creatinine.
We measured bilirubin interference with the kinetic Jaffé method for serum creatinine. Both pooled sera with added bilirubin and icteric patients' sera were used and results with of which gave more nearly "true" values.
متن کاملEvaluation of an enzymatic method for determining creatinine in plasma.
An enzymatic kit method for the determination of plasma creatinine was optimised for use with a centrifugal analyser and its performance characteristics and practicability compared with an end point and a kinetic Jaffé-based method. The enzymatic method exhibited several advantages over Jaffé-based methods--namely, smaller sample size, rapid sample throughput (200 per hour), and improved specif...
متن کاملNew kinetic method for creatinine measurement now automated in the Cobas Fara centrifugal analyzer.
A new kinetic method to determine creatimne in serum by using an alkaline 3,5-dinitrobenzoate solution was recently reported (1). Interference from pseudo-creatinine chromogens, bilirubin, or triacylglycerols was negligible. The results obtained with this kinetic spectrophotometric assay, performed with an Eppendorf ACP 5040 analyzer, agreed well with the procedure of Jaff#{233}, as modified by...
